Best evidence for GrimAge (Lu 2019) for mortality prediction. Individual test-retest reproducibility is poor for older clocks (Horvath v1); newer clocks (DunedinPACE, GrimAge2) better but still noisy — single-point test results can mislead. Use population-level changes over time or intervention-paired measurements, not single absolute values.
Mechanism
DNA methylation patterns at specific CpG sites correlate with biological age and mortality; GrimAge (Lu 2019) additionally incorporates methylation-estimated proteins and smoking; multiple commercial offerings (TruDiagnostic, MyDNAge, Elysium Index)
